چکیده | ||
پژوهش حاضر، برای بررسی پاسخهای فیزیولوژیک و دفاعی ارقام سورگوم قرمز و جارویی نسبت به تنش شوری در شرایط کشت درون شیشه طراحی شد. در ابتدا بذرهای ارقام گیاه سورگوم در محیطکشت MS دارای غلظتهای0، 50، 100 و 150 میلیمولار NaCl در شرایط درون شیشهای کشت شدند. پس از 2 هفته، اثر تنش شوری بر درصد جوانهزنی، شاخصهای رشد، آنتوسیانین کل، فلاونوئیدها، قندهای احیاءکننده، پرولین، یونهای Na+، K+ و Ca2+، پروتئین محلول کل، فعالیت آنزیمهای کاتالاز و آسکوربات پراکسیداز بررسی شد. در پژوهش حاضر، دو رقم قرمز و جارویی، که بهترتیب ارقام حساس و مقاوم نسبت به شوری هستند براساس درصد جوانهزنی و شاخصهای رشد، بررسی شدند. مقدار رنگیزههای فتوسنتزی (کلروفیل کل و کاروتنوئید) و آنتوسیانین با افزایش مقدار نمک در هر دو رقم کاهش یافت، درحالیکه میزان فلاونوئیدها در سه طول موج270، 300 و 330 نانومتر افزایش یافتند. با افزایش شوری میزان پرولین، قند و پروتئین در بافتهای ریشه و ساقة هر دو رقم افزایش پیدا کردند. محتوای یون سدیم در ریشة هر دو رقم و ساقة رقم قرمز افزایش یافت؛ ولی در ساقة رقم جارویی تفاوت معنیداری نشان نداد. بهطورکلی، پتاسیم در تنش شوری کاهش یافت. نسبت سدیم به پتاسیم در ریشة هر دو رقم و ساقة رقم قرمز افزایش نشان داد؛ درحالیکه نسبت سدیم به پتاسیم در ساقة رقم جارویی اثر معنیدار نشان نداد. شوری اثر معنیداری بر میزان یون کلسیم در ریشة دو رقم نداشت. در حالیکه میزان کلسیم در ساقة هر دو رقم افزایش یافت. فعالیت آنزیم CAT در اندام هوایی و ریشة هر دو رقم افزایش یافت؛ ولی فعالیت APX در رقم جارویی افزایش و در رقم قرمز کاهش معنیدار را نشان داد. | ||
