چکیده

Genus Euphlyctis includes two subgenera, Euphlyctis and Phrynoderma, and more than ten lineages in Southeast Asia. In recent studies, skittering frogs of the genus Euphlyctis have been taxonomically revised. Moreover, some new cryptic species have been identified and described through molecular and morphological approaches. Based on the latest taxonomic revision, related species have been placed into two subgenera, and the status of E. cyanophlyctis (Schneider, 1799) has also been revised. In this study, we aim to elucidate the taxonomic status of the only member of the genus Euphlyctis in Iran, distributed in the southeast of Iran. Additionally, the phylogenetic relationship and genetic divergence of Iran's skittering frogs have also been re-evaluated and discussed. In this study, 32 sequences of mitochondrial 16S rRNA, including 492 bp from Iranian skittering frogs, and 284 sequences from GenBank have been analyzed to clarify the phylogenetic relationships and genetic divergence among populations of the species in Iran and other Asian countries. Phylogenetic analyses using BEAST software revealed 13 clades and subclades within the species. Skittering frogs from Iran were placed in the clade that includes specimens from Pakistan, Nepal, Bangladesh, India, and a topotype of E. adolfi, receiving high support (PP 1.00). In addition, this clade was divided into 3 separate lineages, each of which is related to geographical isolations. Despite molecular analyses, morphological features of skittering frogs from southeast Iran demonstrated variations in morphological characters, resulting in complexity and misleading nomenclature.

Introduction
Genus Euphlyctis belongs to the family Dicroglossidae which includes two subgenera Euphlyctis and Phrynoderma and more than ten lineages in the Southeast Asia (Dubois et al., 2021; Dufresnes et al., 2022; Forest, 2024). Subgenus Euphlyctis consists of E. ehrenbergii, E. jaladhara, E. adolfi, and E. cyanophlyctis distributed over south Asia and E. karaavali, E. hexadactyla, E. aloysii, and E. kerala were included in subgenus Phrynoderma living in the south India, Bangladesh, and Sri Lanka (Dinesh et al., 2021, 2022; Dufresnes et al., 2022). The skittering frogs of Iran can be found in the southeast (Sistan and Baluchestan, and Hormozgan) and were first recognized as Rana cyanophlyctis Boulenger, 1920. Baluch and Kami (1994) mentioned them as R. cyanophlyctis seistanica. Later, it has been revised as E. adolfi (Dufresnes et al., 2022); however, Dinesh et al., (2021) recognized E. adolfi (cyanophlyctis) seistanica as a separate species. In this study, we investigated the phylogenetic relationships and genetic divergence among populations of the species of the genus Euphlyctis in the Southeast Asia.

Materials and methods
A total of 45 individuals of skittering frog (E. cyanophlyctis) were collected from nine localities of Sistan and Baluchestan Province, southeast Iran. Of these, 32 tissue samples from 45 specimens were used for molecular study. Total DNA was extracted from muscle tissue using the phenol/chloroform (Sambrook et al., 1989) and mitochondrial 16S rRNA was amplified using 16SL and 16SH primers (Palumbi, 1991). The final data matrix consisted of 316 Euphlyctis sequences (including 32 sequences from this study and 284 sequences retrieved from GenBank), Fejervarya cancrivora (AB070738) and two Hoplobatrachus tigerinus (AB272594) were used as outgroups. Phylogenetic analysis was conducted using the Bayesian method implemented in BEAST v2.2.1 (Bouckaert et al., 2014) with the GTR+I+G model. All analyses were run in the CIPRES Science Gateway (Miller et al., 2010). The genetic divergences between and within lineages were computed using the p-distance method in Mega.

Research Findings
The Bayesian tree based on 316 Euphlyctis sequences revealed 13 clades and subclades with a posterior probability (PP) of ≥0.70 in South Asia. Clade A includes specimens from Iran, Pakistan, India, Bangladesh, and Nepal. This clade was separated into three subclades (A1, A2, and A3) with high PP (1.00). The specimens from Iran and Pakistan were included in subclade A1. The genetic distances based on p-distance ranged from 1.4% to 1.6% between lineages and 0.2% to 0.3% within subclades. In addition, morphological comparisons of the specimens showed high color polymorphism in the southeast of Iran.

Discussion of Results and Conclusions
Iran's skittering frogs form a clade with specimens from Pakistan, India, Nepal, Bangladesh, and the topotypes of E. adolfi (Clade A), indicating that this clade should be assigned to E. adolfi based on the priority rule. Subclade A1 includes specimens from South Iran and South and Central Pakistan, where two subspecies, E. a. seistanica (Nikolskii, 1899) from Hamoon Saburi in North Sistan, Iran, and E. a. microspinulata Khan, 1997 from Khuzdar, Pakistan, have been described. Considering the close proximity of the sampling site of this study (Karvandr Khash) to Hamoon Saburi (about 200 to 300 km), it can be deduced that Iran's skittering frogs are related to the geographical isolate E. a. seistanica, and E. a. microspinulata is its junior synonym, as supported by morphometric and molecular characters. However, molecular and morphological investigation on the type and paratype specimens is needed to elucidate the taxonomic status of Iran's skittering frogs.

مقدمه

قورباغه‌های جنس Euphlyctis Fitzinger, 1843 متعلق به خانواده Dicroglossidae دارای دو زیرجنس Euphlyctis و Phrynoderma است که با بیش از ده دودمان واگرا در جنوب آسیا پراکنده شده‌اند (Dubois et al., 2021; Dufresnes et al., 2022; Forest, 2024). زیرجنس Euphlyctis شامل چهار گونه E. ehrenbergii، E. jaladhara، E. adolfi و E. cyanophlyctis است که در سرتاسر جنوب آسیا و مناطق مجاور آن گسترش دارند(Khajeh et al., 2014; Al-Qahtani & Amer, 2019; Ali et al., 2020; Dinesh et al., 2021; Dufresnes et al., 2022). اعضای زیرجنس Phrynoderma شامل آرایه‌های E. karaavali، E. hexadactyla، E. aloysii و E. keralaو چند آرایه توصیف‌نشده است که در جنوب هند، بنگلادش و سری‌لانکا پراکنش دارند (Dinesh et al., 2021, 2022; Dufresnes et al., 2022).

با ظهور روش‌های نوین و استفاده از داده‌های مولکولی، آرایه‌شناسی و تبارزایی آرایه‌های جنس Euphlyctis به‌ویژه قورباغه‌های مردابی بلوچی ایران بازنگری شده است. تا همین اواخر بیشتر دودمان‌های این جنس به‌دلیل شباهت‌های ریختی تنها در دو گروه گونه‌ای E. cyanophlyctis و E. hexadactyla (Lesson, 1834) طبقه‌بندی می‌شدند (Alam et al., 2008; Khajeh et al., 2014; Howlader et al., 2015; Al-Qahtani & Amer, 2019)؛ اما استفاده از داده‌های مولکولی به‌ویژه ژن 16S rRNA میتوکندری تعداد زیادی نمونه، دودمان‌ها و کلادهای جدیدی در این دو گروه گونه‌ای نمایان کرد که منجر به تغییرات گسترده در آرایه‌شناسی و تبارزایی این جنس شد (Joshy et al., 2009; Howlader et al., 2015; Dinesh et al., 2021, 2022; Dufresnes et al., 2022)؛ به‌طوری‌که تعداد گونه‌های این جنس از پنج گونه به بیش از هشت گونه افزایش یافت (Dufresnes et al., 2022; Frost, 2024).

قورباغه مردابی بلوچی در جنوب ایران در استان‌های سیستان‌وبلوچستان و هرمزگان پراکنش دارد و پیش از این براساس داده‌های ریختی و مولکولی در گروه گونه‌ای E. cyanophlyctis (Schneider, 1799) قرار می‌گرفت (Rastegar-Pouyani et al., 2008; Khajeh et al., 2014)؛ اما داده‌های مولکولی اخیر با استفاده از تعداد زیادی نمونه‌ (286 نمونه) تقریباً از تمام دامنه پراکنش گونه، این گروه گونه‌ای را به چند دودمان مجزا تقسیم کرده و مطابق آن قورباغه‌های مردابی بلوچی ایران در دودمانی با عنوان E. adolfi (Günther, 1860) قرار گرفته‌اند (Dufresnes et al., 2022). قورباغه‌های مردابی بلوچی ایران هاپلوگروهی با نمونه‌هایی از پاکستان و شمال‌غرب هند تشکیل می‌دهند که براساس تحلیل‌های ژنتیک جمعیت طی چند ده هزار سال اخیر فرایند گسترش جمعیتی را تجربه کردند (Dufresnes et al., 2022)؛ بنابراین، استفاده از داده‌های مولکولی آرایه‌شناسی و تبارزایی قورباغه‌های مردابی بلوچی ایران را دچار تغییرات گسترده‌ای کرده است که نیازمند بررسی نمونه‌های بیشتری به‌ویژه از محل تایپ برخی زیرگونه‌های توصیفی از جنوب‌شرق ایران است (Khatiwada et al., 2021; Dinesh et al., 2021; Dufresnes et al., 2022).

جمعیت‌های این گونه در جنوب ایران ابتدا به‌عنوان Rana cyanophlyctis Boulenger, 1920 (خانواده Ranidae) در نظر گرفته شده بود (Anderson, 1963; Boulenger, 1920). سپس Nikolskii (1899) براساس اندازه پرده صماخ و طول پوزه، زیرگونه R. cyanophlyctis seistanica (Nikolskii, 1899) را از مرز ایران، افغانستان و پاکستان در محل تایپ یعنی نیزار سیستان در جنوب‌شرق ایران توصیف کرد؛ اما جمعیت‌های این گونه در جنوب ایران توسط بلوچ و کمی (1994) به‌عنوان زیرگونه Rana cyanophlyctis cyanophlyctis توصیف شدند و زیرگونه R. cyanophlyctis seistanica به‌عنوان همنام متأخر این زیرگونه در نظر گرفته شد؛ اما در چک‌لیست‌های دوزیستان ایران (Rastegar-Pouyani et al., 2008; Safaei-Mahroo et al., 2015) و مطالعه مولکولی Khajeh et al. (2104)، جمعیت‌های این گونه به‌عنوان E. cyanophlyctis در نظر گرفته شدند. علاوه بر این، Khan (1997) زیرگونه جدیدی به نام E. c. microspinulata Khan, 1997 از خوزدر بلوچستان (Khuzdar, Balochistan) در غرب پاکستان توصیف کرد که با توجه به حضور E. adolfi در شمال و جنوب بلوچستان (Dufresnes et al., 2022) احتمال همنامی این زیرگونه با سایر آرایه‌های آن وجود دارد.

نتایج Khajeh et al. (2014) براساس داده‌های ژن 16S میتوکندری 32 نمونه بیان‌کنندة واگرایی بسیار جزئی جمعیت‌های این گونه در جنوب‌شرق ایران بوده‌اند که دودمانی مجزا به‌عنوان گروه خواهری دودمان بنگلادش در گروه گونه‌ای E. cyanophlyctis تشکیل دادند. همچنین، مطالعه Dufresnes et al. (2022) با استفاده از داده‌های ریختی و مولکولی نشان داد جمعیت‌های ایران در زیرجنس Euphlyctis و به گونه E. adolfi تعلق دارند و ازنظر ژنتیکی از نمونه‌های تایپ E. cyanophlyctis متمایز هستند. Dinesh et al. (2021) براساس صفات ریختی و مولکولی E. adolfi (cyanophlyctis) seistanica را به‌عنوان گونه مجزا در نظر گرفته است و آرایه E. kalasgramensis Howlader, Nair, Gopalan, and Merilä, 2015 را براساس قانون حق تقدم با آن همنام کردند. با این حال، دربارة اعتبار آرایه‌شناختی هر دو آرایه E. a. (cyanophlyctis) seistanica و E. a.(cyanophlyctis) microspinulata هنوز تردید وجود دارد و به مطالعه نمونه‌هایی از محل تایپ این دو نیاز است.

قورباغه مردابی بلوچی در جنوب‌شرق ایران در دریاچه هامون، سدها و برکه‌های محلی یافت می‌شود. بسیاری از برکه‌های این منطقه به‌صورت فصلی به‌دنبال باران‌های فصلی و رهاسازی آب در رودخانه هلمند آبگیری می‌شوند. در این ناحیه میزان بارندگی، کم و تفاوت درجه حرارت شب و روز بسیار بالا است (Mohammadi et al., 2015; Mohaymeni et al., 2022)؛ ازاین‌رو، زمین‌های خشک این منطقه عمدتاً توسط رودخانه هلمند و دریاچه هامون آبیاری می‌شوند (Whitney, 2006). با این حال، طی سال‌های اخیر به‌دلیل میزان بارندگی اندک و جلوگیری دولت افغانستان از ورود آب به رودخانه هلمند، تعداد و تنوع دوزیستان این منطقه به شدت کاهش یافته است (Mohammadi et al., 2015) و تنها در برخی زیستگاه‌ها با آب مداوم مانند مناطقی از رود هلمند جمعیت‌هایی از قورباغه مردابی بلوچی یافت می‌شود (Khajeh et al., 2014)؛ ازاین‌رو، طی این مطالعه با توجه به تغییرات آرایه‌شناختی گسترده جنس Euphlyctis و نیز تغییرات اقلیمی درخور توجه در جنوب‌شرق ایران، وضعیت آرایه‌شناسی و تبارزایی جمعیت‌های قورباغه بلوچی با استفاده از داده‌های ژن 16S rRNA میتوکندری بازنگری شده است. علاوه ‌بر این ویژگی‌های ریختی و تغییرات دامنه پراکنش این گونه در جنوب‌شرق ایران در پاسخ به خشکسالی‌های اخیر بررسی شده‌اند.

مواد و روش‌ها

طی این تحقیق 45 نمونه قورباغه بلوچی از 9 منطقه مختلف در جنوب‌شرق ایران با استفاده از دست و تور دستی جمع‌آوری شدند (جدول 1 و شکل 1). انتخاب مناطق نمونه‌برداری مطابق شرایط اقلیمی منطقه یعنی وجود رودخانه، برکه و باتلاق صورت گرفت. نمونه‌ها براساس صفات ریختی مطابق منابع قابل دسترس شناسایی شدند (Baluch & Kami, 1995; Khan, 1997; Safaei-Mahroo et al., 2023) و ازنظر ویژگی‌هایی مانند رنگ بدن، شکل خال‌های پشتی و شکمی و وجود پرده میان انگشتان پا بررسی شدند. سپس از ماهیچه ران آنها نمونه بافتی تهیه شد و برای مطالعات مولکولی داخل اتانول 96 درصد قرار داده شدند. تمام نمونه‌ها و بافت‌ها در آزمایشگاه جانورشناسی دانشگاه سراوان نگهداری می‌شوند. برای مطالعه نمونه‌های جمع‌آوری‌شده، کد اخلاق از مدیریت پژوهشی مجتمع عالی سراوان به شماره IR.Saravan.Com.1390.031 دریافت شد.

استخراج DNA و PCR

از 45 نمونه جمع‌آوری‌شده، 32 نمونه مطابق ویژگی‌های ریختی برای مطالعات مولکولی انتخاب شدند. استخراج DNA از بافت ماهیچه‌ای به روش نمکی طبق دستورالعمل فنول - کلروفرم (phenol/chloroform) صورت گرفت (Sambrook et al., 1989). صحت DNA استخراج‌شده با استفاده از نانودراپ و ژل آگارز سنجیده شد و نمونه‌هایی با غلظت DNA بالاتر از 50 نانوگرم / میکرولیتر استفاده شدند. سپس واکنش زنجیره‌ای پلی‌مراز (PCR) با استفاده از آغازگر جلورونده 16SL (5´-CGCCTGTTTATCAAAAACAT-3´) و آغازگر معکوس 16SH (5´-CCGGTCTGAACTCAGATCACG-3´) مطابق پروتوکل Palumbi (1991) برای تکثیر ژن 16S rRNA میتوکندری صورت گرفت. محصولات PCR مثبت برای تعیین توالی به شرکت ماکروژن کره جنوبی ارسال شدند. تعیین توالی با استفاده از دستگاه ABI Prism 3700 صورت گرفت که در بانک ژن با شماره‌های KF992800 تا Kf992833 در دسترس هستند (جدول ۲).

تحلیل‌های تبارزایی

ابتدا توالی‌های ژن 16S میتوکندری این مطالعه با استفاده از برنامه Clustal W (Thompson et al., 1994) در نرم‌افزار Bioedit v.7.0.9 ویرایش شدند(Hall, 1999) . سپس 284 توالی مربوط به جنس Euphlyctis از بانک ژن، دریافت و به توالی‌های این مطالعه اضافه شدند. در مجموع، 316 توالی (32 توالی این مطالعه + 284 توالی از بانک ژن) با نرم‌افزار Bioedit هم‌تراز شدند و سپس فایل نکسوس این 316 توالی به اضافه دو توالی Hoplobatrachus tigerinus (AB272594) و cancrivorus Fejervarya (AB070738) به‌عنوان برون‌گروه با استفاده از نرم‌افزار Mega v.11 (Tamura et al., 2021) ساخته شد. از نرم‌افزار jModelTest (Darriba et al., 2012) برای انتخاب بهترین مدل تکامل مولکولی استفاده شد که براساس معیار Bayesian Information Criterion (BIC) (Schwarz, 1978) مدل تکاملی GTR+I+G انتخاب شد.

درنهایت، فایل XML داده‌ها با استفاده از نرم‌افزار BEAUti با استفاده از مدل relaxed log-normal در بسته نرم‌افزاری BEAST v.2.2.1 (Bouckaert et al., 2014) ساخته شد و تحلیل تبارزادی به روش بیزین Bayesian inference (BI) برای این 318 توالی توسط نرم‌افزار BEAST صورت گرفت. تحلیل بیزین با ۱۰۰ میلیون تکرار با استفاده از روش Markov Chain Monte Carlo (MCMC) اجرا شد و از هر ۵۰۰۰ تکرار پارامترها و توپولوژی درخت مولکولی نمونه‌برداری شدند. سپس ۲۵ درصد اول درختان مولکولی برای افزایش اطمینان به‌عنوان درخت‌های بی‌اعتبار به‌عنوان Burn-in کنار گذاشته شدند. از درخت‌های باقی‌مانده درخت اجماع با مقادیر تأییدی بیش از 70 درصد و بیشترین میانگین توسط نرم‌افزارTreeAnnotator v.2.2.1 محاسبه شد؛ درنهایت، درخت با نرم‌افزار Figtree v1.4.3 ترسیم شد (Rambaut, 2016). این تحلیل‌ها در وبگاه CIPRES Science Gateway انجام شدند (Miller et al., 2010). همچنین، فاصله ژنتیکی ناشی از جهش و تغییر نوکلئوتیدی درون و بین دودمان‌ها توسط مدل تکاملی p-distance در نرم‌افزار Mega محاسبه شد (Kimura, 1980).

تحلیل تبارزایی به روش بیزین براساس 492 جفت‌باز از ژن 16S rRNA میتوکندری برای 316 توالی از جنس Euphlyctis صورت گرفت. درخت مولکولی بیزین نشان داد این 316 توالی در 13 کلاد و زیرکلاد قرار می‌گیرند (شکل 2) که برخی از آنها در ارتباط با دودمان‌های جدیدی هستند که نیاز به توصیف یا بازنگری دارند. براساس این داده‌ها کلاد A نمونه‌هایی از ایران (هرمزگان و سیستان‌وبلوچستان)، پاکستان، هند، بنگلادش و نپال را شامل می‌شود که با مقدار تأییدی بسیار بالا حمایت می‌شود (PP 1.00). این کلاد به سه زیرکلاد شامل A1، A2 و A3 تقسیم می‌شود که در زیرکلاد A1 نمونه‌های ایران همراه با پاکستان و شمال‌غرب هند با مقادیر تأییدی بالا (PP 1.00) قرار می‌گیرند. در زیرکلاد A2 نمونه‌هایی از پاکستان و در زیرکلاد A3 نمونه‌هایی از پاکستان، هند، بنگلادش و نپال قرار می‌گیرند که با مقادیر تأییدی بالا (PP ≥ 0.99) حمایت می‌شوند. با این حال، رابطه نزدیک زیرکلاد A1 و A2 با مقادیر تأییدی حمایت نمی‌شود (PP 0.70)؛ اما رابطه خواهری زیرکلادهای A1 و A2 با A3 به‌طور کامل حمایت می‌شود‌ (PP 1.00). همچنین کلاد A با کلاد حاوی نمونه‌های E. jaladhara از هند با مقادیر تأییدی متوسط (PP 0.76) کلاد خواهری تشکیل می‌دهد. علاوه بر این، فاصله ژنتیکی براساس مدل p-distance بین دودمان‌های کلاد A نشان داد فاصله مولکولی دودمان A1 شامل نمونه‌های قورباغه‌های بلوچی ایران با A2 6/1 درصد و با A3 4/1درصد است. همچنین، فاصله ژنتیکی درون دودمان‌ها بین 2/0 و 3/0 درصد بود (جدول ۳).

ریختی

بررسی شکل ظاهری نمونه‌های قورباغه بلوچی از مناطق مختلف جنوب ایران برخلاف نتایج مولکولی نشان داد این نمونه‌ها ازنظر رنگ بدن و شکل خال‌های شکمی و پشتی دارای تغییرات زیادی هستند و ازنظر صفات ظاهری چندریختی زیادی را نشان می‌دهند (شکل 3). رنگ سطح پشتی بدن از زیتونی روشن تا قهوه‌ای تیره و رنگ سطح شکمی از شیری تا زرد متغیر بود. قورباغه‌های بلوچی ازنظر خال‌های پشتی و شکمی نیز تغییرات زیادی را نشان می‌دهند؛ به‌طوری‌که در برخی این خال‌ها بسیار ریز، در برخی دیگر درشت و حتی برخی فاقد خال بودند.

بحث

مطالعه و شناسایی گونه‌های هم‌ریخت یا دارای تفاوت‌های ریختی اندک (نهان گونه‌ها)، به کمک روش‌های مولکولی به یک مهم در مطالعات تنوع زیستی تبدیل شده است (Dinesh et al., 2022). درواقع، استفاده از داده‌های مولکولی منجر به بازنگری گسترده بسیاری از همتافت گونه‌های دوزیستان شده است و به موجب آن بسیاری از آرایه‌ها یا همنام شده‌اند یا آرایه‌شناسی آنها تغییر یافته است (Freilich et al., 2014; Wielstra et al., 2018; Scherz et al., 2019; Dinesh et al., 2022; Dufresnes et al., 2022). E. cyanophlyctis از همتافت گونه‌هایی با دامنه پراکنش گسترده در جنوب و جنوب‌شرق آسیا است که آرایه‌شناسی آن اخیراً دچار تغییرات گسترده‌ای شده است. مطابق نتایج پژوهش حاضر و مطالعه Dufresnes et al. (2022) نمونه‌های این همتافت گونه از ایران، پاکستان، هند، بنگلادش و نپال با استفاده از داده‌های ژن 16S میتوکندری به بیش از 13 دودمان واگرا با مقادیر تأییدی نسبتاً بالا تقسیم می‌شود که هریک می‌تواند به‌عنوان گونه یا زیرگونه مجزایی در نظر گرفته شود؛ با این حال، برای تأیید این موضوع نیاز به داده‌های بیشتری ازجمله مطالعه آرایه‌شناسی و تبارزایی آنها با استفاده از ژن‌های هسته‌ای و توالی ژنوم کامل است.

نمونه‌های قورباغه بلوچی جنوب ایران همراه با نمونه‌های پاکستان، هند، نپال و بنگلادش در کلاد A قرار می‌گیرد که با توجه به قانون حق تقدم و حضور توپوتایپ‌های E. adolfi در این کلاد به‌عنوان این گونه در نظر گرفته می‌شود؛ ازاین‌رو، نمونه‌های ایران به E. adolfi تعلق دارند و حضور E. cyanophlyctis در ایران باید بررسی شود. با وجود این، دو گونه E. cyanophlyctis و E. adolfi ازنظر صفات ریختی و اندازشی بسیار شبیه به هم بوده‌اند و از همدیگر تفکیک‌پذیر نیستند (Dufresnes et al., 2022). هرچند تفاوت‌هایی ازنظر خال‌های شکمی و الگوهای رنگ پشت بدن و نیز اندازه بدن برای تفکیک این دو گونه ارائه شده است (Howlader et al., 2015)؛ اما بررسی نمونه‌های این مطالعه بیان‌کنندة چندریختی گسترده ازنظر رنگ بین مناطق مختلف است؛ ازاین‌رو، مقایسه این دو با استفاده از نمونه‌های محدود، با توجه به تغییرات گسترده رنگ و خال‌های بدن، معیار خوبی برای شناسایی و مقایسه این گونه‌های خویشاوند نیست.

واگرایی کلاد A به سه زیرکلاد A1، A2 و A3 با فاصله ژنتیکی حدود 4/1 تا 6/1 درصد نشان می‌دهد می‌توان هریک از این زیرکلادها را به زیرگونه‌های توصیفی از دامنه پراکنش آنها اختصاص داد. در زیرکلاد A1 نمونه‌های جنوب ایران همراه با نمونه‌هایی از جنوب و مرکز پاکستان قرار دارند که تاکنون دو زیرگونه E. a seistanica (Nikolskii, 1899) از نیزار هامون صابوری در شمال سیستان ایران و زیرگونه E. a. microspinulata Khan, 1997 از خوزدر در جنوب بلوچستان پاکستان توصیف شده است. با توجه به اینکه فاصله نیزار هامون صابوری تا نزدیک‌ترین منطقه نمونه‌برداری در این مطالعه، یعنی کارواندر خاش، حدود 200 تا 300 کیلومتر است، می‌توان این زیرکلاد را تحت E. a seistanica (Nikolskii, 1899) در نظر گرفت و با توجه به قانون حق تقدم زیرگونه E. a. microspinulata Khan, 1997 را به‌عنوان همنام مؤخر آن در نظر گرفت. علاوه بر این، شباهت جمعیت‌های قورباغه مردابی بلوچی جنوب ایران ازنظر صفات اندازشی و مولکولی (Khajeh et al., 2014) نیز از این پیشنهاد حمایت می‌کند. با این حال، طی سال‌های اخیر هامون صابوری به شدت خشک شده است (Mohammadi et al., 2015) و با توجه به مراجعه چندباره نویسندگان این پژوهش به این منطقه قورباغه‌ای در آن یافت نشد. گرچه برای تشخیص دقیق وضعیت آرایه‌شناختی این زیرگونه نیاز به مطالعه مولکولی نمونه تایپ یا نمونه‌های پاراتایپ است.

به نظر می‌رسد خشکسالی‌های گسترده و پیامد آن، ایجاد دشت‌های خشک و لم‌یزرع در دوره نئوژن (Whitney, 2006) به‌عنوان یک مانع جغرافیایی عمل کرده‌اند و سبب جدایی و واگرایی دودمان‌های این گونه در حوضه هلمند از دودمان‌های شبه‌قاره هند شده‌اند (Khajeh et al., 2014; Dinesh et al., 2021). همین مسئله در واگرایی دودمان‌های جنس Laudakia و همتافت گونه Eremias persica نیز تأثیر گذاشته است (Rastegar Pouyani et al., 2010). خشکسالی‌های اخیر منطقه سبب شده‌اند جمعیت‌های قورباغه بلوچی در جنوب ایران، به‌ویژه در شمال استان‌های هرمزگان و سیستان‌وبلوچستان، به شدت کاهش یابند یا به‌طور کل ناپدید شوند.
