چکیده

The honey bee is one of the most economically important insect species due to its crucial role in the pollination of agricultural and non-agricultural plants, as well as its direct product contributions. The western honey bee, Apis mellifera, now considered a cosmopolitan species due to its widespread distribution by humans, originated in Africa, Europe, and West Asia. This study aimed to evaluate the diversity of this bee in parts of northwest Iran. Worker bees were collected from six study stations in West Azerbaijan and East Azerbaijan provinces and assessed using the geometric morphometric method. Changes in the size and shape of the forewings and hindwings were investigated using 16 homologous landmarks at the bifurcation of the wings. Regarding the size of the forewings and hindwings, the Chahar Borj population of West Azerbaijan had the largest, while the Malekan population of East Azerbaijan had the smallest wings. Significant differences were observed between all the study stations of East Azerbaijan and those of West Azerbaijan. In terms of forewing shape, significant differences were found in all pairwise comparisons except between the populations of Sahand Mountain and Kordeh Deh village. However, for the hindwing, significant differences were observed in only three pairwise comparisons. Additionally, the regression analysis revealed that, unlike the hindwing, changes in the forewing's size and shape are interdependent, with allometry also being observed. This study demonstrates that despite the geographical proximity of the study stations, significant diversity exists in the bee populations.
Introduction
Bees are insects with a lineage dating back 120 million years. Honey bees include 10 species belonging to the genus Apis, of which the species Apis mellifera has become a cosmopolitan species with the help of humans. Honey bees produce valuable products for humans and play a crucial role in crop pollination. In recent years, this species has experienced a global decline due to factors such as habitat destruction, pollution, pesticides, and diseases. One aspect that can aid in the adaptation of bees and better equip them to face these challenges is genetic diversity. In this study, the morphological diversity of honey bees, which can sometimes reflect genetic diversity, was investigated in areas of northwest Iran using the geometric morphometric method. This approach is expected to provide insights into the adaptability and resilience of honey bee populations in these regions.

Materials and Methods
A total of 218 bees were collected from 6 study stations located in West Azerbaijan and East Azerbaijan provinces during the spring and summer of 2022 with the cooperation of beekeepers and were transferred to the laboratory and placed in ethanol. Microscopic slides of the right forewing and hindwing of 180 bees were prepared and were photographed using a stereomicroscope equipped with a camera. The number of 16 landmarks in the forewing and 5 landmarks in the hindwing were selected at the bifurcation of the wings and were landmarked using tpsDig2 software. The coordinates of the landmarks were also saved. Then, these coordinates were statistically analyzed in different pieces of software in order to detect possible differences in the shape and size of the fore and hindwings in different populations.
Research Findings
In terms of the size of the forewings and hindwings, comparisons between the populations revealed diversity. The average centroid size of the forewings and hindwings indicated that the Chahar Borj honey bee population had the largest wings, while the Malekan honey bee population had the smallest. A one-way non-parametric Kruskal-Wallis Analysis of Variance, along with its post hoc tests on the centroid size data of the forewings and hindwings, revealed significant differences among most of the populations. Regarding the general shape of the forewings and hindwings, various analyses, including Principal Component Analysis, Canonical Variate Analysis, and Multivariate Analysis of Variance, highlighted diversity and significant differences between the populations. Cluster Analysis of the populations based on forewing and hindwing data further divided these populations into three distinct clusters. In examining the relationship between wing size and shape, Regression Analysis indicated the presence of allometry solely in the forewing.

Discussion of Results and Conclusion
Economically, honey bees are one of the most important insect species. Both morphological and molecular characteristics are employed to evaluate their diversity and to differentiate between populations and species. In recent years, geometric morphometry has been increasingly utilized to distinguish between species, subspecies, and populations. In this study, geometric morphometrics was applied to assess the diversity of the forewings and hindwings of bees. The results revealed that despite the sampled populations being located in a relatively small geographical area, they exhibited a considerable amount of diversity. This finding is significant because a lack of population diversity in honey bees renders them more vulnerable to pests, diseases, and the impacts of climate change. The study underscores the importance of understanding and preserving the genetic and morphological diversity within bee populations to enhance their resilience against environmental challenges.

مقدمه

زنبورها از زمانی که دایناسورها در بیش از 120 میلیون سال پیش روی زمین راه می‌‌رفتند، پرواز و گیاهان گلدار را گرده‌‌افشانی می‌‌کرده‌‌اند. در طول صد میلیون سال گذشته، زنبورها و گیاهان گلدار به گونه‌‌ای تکامل یافته‌‌اند که بدون یکدیگر روی زمین یافت نمی‌‌شوند. زنبورها علاوه‌‌بر نقش مهمی که در گرده‌‌افشانی و تأمین امنیت غذایی دارند، محصولات باارزش دیگری مانند عسل، موم، بره‌‌موم و ژل رویال را در اختیار ما قرار می‌‌دهند که در ساخت سایر محصولات باارزش مانند داروها، شربت‌‌ها، شمع‌‌ها، مرهم‌‌ها، پمادها، محصولات آرایشی، محصولات مرتبط با مو و غیره استفاده می‌‌شوند(Cardinal & Danforth, 2013; Kane & Faux, 2021) .

به‌‌طور کلی، آنچه که زنبور عسل نامیده می‌‌شود شامل 10 گونه متعلق به جنس Apis، خانوادۀ Apidae و راستۀ Hymenoptera است. زنبور عسل غربی، Apis mellifera، به‌‌طور طبیعی در غرب آسیا، اروپا و آفریقا پراکنش دارد (Engel, 1999; Arias & Sheppard, 2005; Han et al., 2012)؛ اما اهلی‌‌سازی و انتقال این گونه توسط انسان، آن را به گونه‌‌ای جهان‌‌وطن تبدیل کرده است که در سراسر جهان به‌‌جز قطب جنوب و بسیاری از جزایر اقیانوسی پراکنده شده است (Ruttner, 1988; Hung et al., 2018). نه گونۀ دیگر Apis به‌‌طور انحصاری در آسیا یافت می‌‌شود (Han et al., 2012). پژوهش‌‌های مورفولوژیک و مولکولی پنج دودمان اصلی تکاملی زنبورهای عسل شامل A (آفریقا) و زیرنژاد آن Z، M (اروپای غربی و شمالی)، C (اروپای شرقی)، O (خاور نزدیک و آسیای مرکزی) و Y (شرق آفریقا و شبه‌‌جزیرۀ عربی) با بیش از 33 زیرگونه (که نژادهای جغرافیایی نیز نامیده می‌‌شود) را نشان می‌دهد (Ruttner, 1988; Requier et al., 2019; Ilyasov et al., 2020). از این دودمان‌‌ها و زیرگونه‌ها، Apis mellifera meda Skorikov 1929b (زنبور عسل ایرانی) و Apis mellifera anatoliaca Maa 1953 (زنبور عسل آناتولی) متعلق به دودمان A (فرعی Z) و همچنین Apis mellifera remipes Gerstäcker1 متعلق به دودمان O، به نظر می‌‌رسد در ایران پراکنش داشته باشند (Ilyasov et al., 2020).

گرده‌افشان‌های وحشی و زنبورهای عسل به‌‌دلیل فعالیت‌های انسانی مانند تخریب زیستگاه، آلودگی، آفت‌کش‌ها و بیماری‌ها کاهشی جهانی را در سطوح مختلف تجربه می‌کنند (Kükrer et al., 2021)؛ علاوه‌‌بر این زنبورهای عسل بومی درنتیجۀ اختلاط ژنتیکی به‌‌دلیل فعالیت‌‌های تجاری، مانند جایگزینی زنبورهای عسل محلی با گونه‌‌های غیر بومی و شیوه‌‌های زنبورداری که کلنی‌‌ها را بین مناطق جغرافیایی جابجا می‌‌کند، از انقراض محلی رنج می‌‌برند (De La Rúa et al., 2009). این چالش‌ها تأثیرات منفی بر جمعیت زنبور عسل دارند و به‌‌طور احتمالی به انقراض آنها کمک می‌کنند؛ اما گاهی اوقات جمعیت‌های بومی به‌‌دلیل ساختار ژنتیکی خود با این چالش‌ها بهتر مواجه می‌شوند (Büchler et al., 2014). تنوع، کلید سازگاری با این شرایط است؛ بنابراین مطالعۀ تنوع ژنتیکی و مورفولوژیکی زنبور عسل و حفظ و نگهداری آن در سطوح مختلف، یعنی سطوح جمعیت و زیرگونه‌ها، برای حفاظت از گونه‌ها و اکوسیستم‌های زنبورهای عسل و خدمات اقتصادی ارائه‌‌شده توسط آنها، اهمیت زیادی دارد (Kükrer et al., 2021).

تعداد مطالعات متمرکز بر جمعیت زنبور عسل ایران رو به افزایش است. در این مطالعات از روش‌‌های مورفولوژیکی (Tahmasebi et al., 1998; Parichehreh Dizji et al., 2017; Boulhasani, et al., 2018; Dadgostar et al., 2020a; Dadgostar, et al., 2020b) و روش‌‌های مولکولی (Rahimi et al., 2016, 2018, 2022; Salehi & Nazemi-Rafie, 2020) برای بررسی ساختار جمعیت‌‌های زنبور عسل ایرانی Apis mellifera meda استفاده شده است. در پژوهش حاضر، از روش مورفومتریک هندسی برای بررسی تفاوت بین جمعیت‌‌های جمع‌‌آوری‌‌شده از مکان‌‌های مختلف استان‌‌های آذربایجان غربی و شرقی استفاده شده است.

مواد و روش‌‌ها

نمونه‌‌برداری و آماده‌‌سازی نمونه‌‌ها

تعداد 218 عدد زنبور کارگر از شش ایستگاه مطالعاتی واقع در استان‌‌های آذربایجان غربی و آذربایجان شرقی (جدول 1) طی بهار و تابستان 1401 با همکاری زنبورداران جمع‌‌آوری، به آزمایشگاه منتقل و در اتانول قرار داده شد. به اذعان زنبورداران، زنبورهای منطقه از نژاد قفقازی هستند. نمونه‌‌ها با تکاندن کادر داخل کندو به داخل کیسۀ پلاستیکی جمع‌‌آوری شد. بال جلو و عقب سمت راستِ 180 زنبور به‌‌دقت جدا شد و روی لام قرار گرفت و بال‌‌های آسیب‌‌دیده در ساخت لام استفاده نشد. بعد از اضافه‌‌کردن یک قطره اتانول روی بال‌‌ها، لامل روی آنها قرار گرفت و اسلاید بال آماده شد. اسلاید‌‌های آماده‌‌شده بلافاصله با استفاده از سیستم تجزیه و تحلیل تصویر متشکل از استریومیکروسکوپ Nikon SMZ-2B شرکت نیکون (Nikon: 9-16, Ohi 3-Chome, Shinagawa-Ku, Tokyo 140, Japan) و دوربین دیجیتال میکروسکوپ شرکت سلسترون (Celestron: 2835 Columbia St. Torrance, California 90503 U.S.A) متصل به رایانه تصویربرداری شد. اسلایدها تا حد امکان در جهت یکسان و مشابه در زیر استریومیکروسکوپ قرار گرفت. تمام تصاویر با درشت‌‌نمایی 6/0 گرفته و تصاویر مربوط به نمونه‌‌های هر جمعیت در پوشه‌‌های جداگانه‌‌ای در رایانه ذخیره شد.

عددی‌‌کردن تصاویر و آماده‌‌سازی داده‌‌ها برای تجزیه و تحلیل آماری

تعداد 16 لندمارک در بال جلو و پنج لندمارک در بال عقب در محل انشعاب رگبال‌‌ها انتخاب شد. لندمارک‌‌گذاری روی تصاویر در محیط نرم‌‌افزار tpsdig2 نسخۀ 64 بیت ویرایش 32/2 (Rohlf, 2015) صورت گرفت و فایل مختصات لندمارک tps برای هر جمعیت ایجاد شد. لندمارک‌‌گذاری با ترتیب یکسانی (شکل 2) در همۀ تصاویر صورت گرفت و تصاویری که به هردلیل فاقد موقعیت هرکدام از لندمارک‌‌ها بود، از لندمارک‌‌گذاری حذف شد. درنهایت، داده‌‌های لندمارک مربوط به تمام جمعیت‌‌ها با ترتیب مشخص‌‌شده در جدول 1 در فایلی ذخیره و آمادۀ تجزیه و تحلیل شد.

تجزیه و تحلیل آماری داده‌‌ها

ماتریس دادۀ کل لندمارک‌‌ها به تفکیک بال جلو و بال عقب وارد نرم‌‌افزار MorphoJ ویرایش 1.07a (Klingenberg, 2011) و برای احتمال وجود داده‌‌های خارج از محدودۀ (Outlier) ناشی از اشتباهات احتمالی در رعایت ترتیب لندمارک‌‌ها در لندمارک‌‌گذاری، ارزیابی شد؛ سپس به‌‌منظور حذف تغییرات غیر شکلی و ترازکردن و انطباق (Superimposition) لندمارک‌‌ها، تحلیل پروکراستی تعمیم‌‌یافته (Generalized Procrustes Analysis) در نرم‌‌افزار MorphoJ و tpsRelw نسخۀ 64 بیت ویرایش 75/1 (Rohlf, 2015) انجام شد. ماتریس اندازۀ مرکزی و ماتریس وزنی محاسبه‌‌شده توسط نرم‌‌افزار tpsRelw برای بال جلو و بال عقب به‌‌طور جداگانه به‌‌منظور تجزیه و تحلیل‌‌های بعدی ذخیره شد. اثر آلومتریک برای ارزیابی اینکه آیا رابطه‌‌ای بین تغییرات شکل و اندازه وجود دارد یا خیر با استفاده از ماتریس وزنی و ماتریس اندازۀ مرکزی و با کمک بستۀ نرم‌‌افزاریtpsRegr نسخۀ 64 بیت ویرایش 50/1 (Rohlf, 2015) محاسبه شد. تجزیه و تحلیل مؤلفه‌‌های اصلی (Principle Component Analysis) به عنوان ابزاری برای بررسی الگوهای تنوع در جمعیت با استفاده از ماتریس واریانس - کوواریانس و تجزیه و تحلیل متغیر کانونی (Canonical Variate Analysis) برای تجزیه و تحلیل و آزمون تفاوت بین جمعیت‌‌ها به ترتیب با استفاده از نرم‌‌افزار MorphoJ و NTSYSpc ویرایش 02/2 (Rohlf, 2000) صورت گرفت. تحلیل واریانس تک‌‌متغیرۀ غیر پارامتری Kruskal-Wallis روی داده‌‌های اندازۀ مرکزی برای تشخیص تفاوت در اندازۀ بال‌‌ها در جمعیت‌‌ها به همراه آزمون تعقیبی (آزمون Dunn’s) برای بررسی تفاوت‌های احتمالی بین هر جفت از جمعیت‌‌های مقایسه‌‌شده انجام شد. تحلیل واریانس چندمتغیرۀ PERMANOVA یک‌‌طرفه روی داده‌‌های مختصات لندمارکی انطباق‌‌یافته با تحلیل پروکراستی برای تشخیص تفاوت در شکل بال‌‌ها در جمعیت‌‌ها توسط نرم‌‌افزار PAST v4.12 (Hammer et al., 2001) با 9999 جایگشت (Permutation) به همراه آزمون تعقیبی Bonferroni-corrected انجام شد. آزمون‌های جایگشت برخلاف نسخه‌های پارامتریک MANOVA به فرضیاتی مانند نرمال‌‌بودن داده‌ها نیاز ندارند. خوشه‌‌بندی یا تحلیل خوشه‌‌ای روی داده‌‌های ماتریس وزنی برای تشخیص ارتباطات فنتیک بین جمعیت‌‌های مطالعه‌‌شده توسط نرم‌‌افزار NTSYSpc انجام شد.

نتایج

تنوع اندازۀ بال جلو و بال عقب

اندازۀ مرکزی که با ریشۀ دوم مجموع مربع فاصلۀ هر لندمارک تا مرکز آرایش فضایی لندمارک‌‌های هر نمونه محاسبه می‌‌شود، به عنوان اندازۀ هندسی هر نمونه معرفی می‌‌شود. میانگین اندازۀ مرکزی در بال جلو، جمعیت زنبورهای عسل چهاربورج را واجد بزرگ‌‌ترین بال‌‌ها و جمعیت زنبورهای عسل ملکان را دارای کوچک‌‌ترین بال‌‌ها نشان داد (تصویر سمت چپ شکل 3)؛ همچنین میانگین اندازۀ مرکزی در بال عقب، جمعیت زنبورهای عسل چهاربورج را واجد بزرگ‌‌ترین بال‌‌ها و جمعیت زنبورهای عسل ملکان را دارای کوچک‌‌ترین بال‌‌ها نشان داد (تصویر سمت راست شکل 3).

تحلیل واریانس یک‌‌طرفه (ANOVA)

تحلیل واریانس یک‌‌طرفه (ANOVA) روشی آماری برای آزمایش این فرضیۀ صفر است که چندین نمونۀ تک‌‌متغیره از جمعیت‌‌های با میانگین یکسان گرفته شده است. فرض می‌‌شود که نمونه‌‌ها نزدیک به توزیع نرمال و دارای واریانس‌‌های مشابهی هستند. اگر تعداد نمونه‌‌های جمعیت‌‌ها برابر باشد، این دو فرض، حیاتی نیست؛ اما اگر مفروضات به‌‌شدت نقض شود، باید به جای آن از تحلیل واریانس یک‌‌طرفۀ غیرپارامتری Kruskal-Wallis استفاده شود. نتایج آزمون نرمال Shapiro-Wilk و آزمون همگنی Levene’s روی داده‌‌های اندازۀ مرکزی بال‌‌های جلو و عقب به‌‌طور جداگانه نشان داد که برخی از جمعیت‌‌ها از توزیع نرمال داده‌‌ها و همگنی واریانس میانگین‌‌ها پیروی نمی‌‌کنند؛ درنتیجه با توجه به نابرابربودن تعداد نمونه‌‌ها در جمعیت‌‌ها، استفاده از تحلیل واریانس یک‌‌طرفۀ معمول ممکن نیست. تجزیه و تحلیل واریانس یک‌‌طرفۀ غیرپارامتری Kruskal-Wallis روی داده‌‌‌های کل اندازۀ مرکزی بال جلو و بال عقب به‌‌طور جداگانه برای بررسی تفاوت‌های احتمالی معنی‌دار بین جمعیت‌‌ها و درون جمعیت‌ها انجام شد و آزمون تعقیبی (آزمون Dunn’s) برای بررسی تفاوت‌های احتمالی بین هر جفت از جمعیت‌‌های مقایسه‌‌شده انجام شد. نتایج تحلیل Kruskal-Walli روی اندازۀ مرکزی نشان داد بین جمعیت‌‌ها ازنظر اندازۀ بال جلو (P=1.536E-29) و بال عقب (P=6.714E-28) اختلاف معنی‌‌دار وجود دارد و نتایج آزمون تعقیبی دان نیز نشان داد که بیشتر جمعیت‌‌های مقایسه‌‌شده با هم اختلاف معنی‌‌دار دارند (جدول 2). از میان جمعیت‌‌های بررسی‌‌شده در رابطه با اندازۀ مرکزی بال جلو، تنها جمعیت‌‌های نقده - چهاربورج، نقده - میاندوآب، چهاربورج - میاندوآب، روستای کرده‌‌ده - کوه سهند، روستای کرده‌‌ده - ملکان و کوه سهند - ملکان فاقد اختلاف معنی‌‌دار بود؛ همچنین از میان جمعیت‌‌های بررسی‌‌شده در رابطه با اندازۀ مرکزی بال عقب، با الگوی مشابهی جمعیت‌‌های یادشدۀ فوق فاقد اختلاف معنی‌‌دار بود (جدول 2).

تنوع شکل بال جلو و بال عقب

تجزیه و تحلیل مؤلفه‌‌های اصلی (PCA)

با تجزیه و تحلیل مؤلفه‌‌های اصلی (PCA) تمام نمونه‌‌ها توسط نرم‌‌افزار MorphoJ، حدود 818/32 درصد تنوع شکل بین نمونه‌‌ها توسط دو مؤلفۀ اول استخراج‌‌شده از ماتریس واریانس - کوواریانس توضیح داده شد (مؤلفۀ اول 192/18 درصد و مؤلفۀ دوم 626/14 درصد)؛ همچنین 817/82 درصد تنوع توسط 10 مؤلفۀ اول از 28 مؤلفه توضیح داده شد. در بال عقب نیز دو مؤلفۀ اول توضیح‌‌دهندۀ حدود 395/57 درصد تنوع شکل بین نمونه‌‌ها بود (مؤلفۀ اول 571/33 درصد و مؤلفۀ دوم 824/23 درصد) و شش مؤلفه، 100 درصد تنوع را توضیح داد. شکل 4 ترسیم پراکندۀ (scatter plot) حاصل از این تحلیل را نشان می‌‌دهد.

تجزیه و تحلیل متغیر کانونی (CVA)

نتایج تحلیل متغیر کانونی روی بال جلو و بال عقب در شکل 5 نشان داده شده است. در این نمودار، صفحه‌‌ها نشان‌‌دهندۀ آرایش فضایی مرجع، ارتفاع میله‌‌ها نشان‌‌دهندۀ فاصلۀ هر جمعیت با آرایش فضایی مرجع و فاصلۀ میله‌‌ها از هم نشان‌‌دهندۀ فاصلۀ جمعیت‌‌ها از یکدیگر است. براساس نتایج در بال جلو، جمعیت‌‌های روستای کرده‌‌ده و ملکان به ترتیب بیشترین و کمترین اختلاف را با آرایش فضایی مرجع نشان داد. طبق نتایج در بال عقب نیز جمعیت‌‌های نقده و ملکان به ترتیب بیشترین و کمترین اختلاف را با آرایش فضایی مرجع نشان داد.

تجزیه و تحلیل چندمتغیرۀ واریانس (MANOVA)

تجزیه و تحلیل چندمتغیرۀ واریانس PERMANOVA که از آزمون جایگشت (Permutation Test) برای تعیین معنی‌‌داری تفاوت‌‌ها استفاده می‌‌کند، روی داده‌‌های مختصات لندمارکی انطباق‌‌یافته با تحلیل پروکراستی توسط نرم‌‌افزار PAST v4.12 با تعداد 9999 جایگشت انجام شد. نتایج این آزمون نشان داد بین جمعیت زنبورها تفاوت معنی‌‌داری ازلحاظ شکل بال جلو (F=5.452, p (same)= 0.0001) و بال عقب (F=1.164, p (same)= 0.0006) وجود داشت. نتایج آزمون تعقیبی نشان داد در بال جلو غیر از جمعیت‌‌های روستای کرده‌‌ده و کوه سهند، بقیۀ مقایسۀ جمعیت‌‌ها اختلاف معنی‌‌دار داشت؛ اما در بال عقب به‌‌جز جمعیت‌‌های روستای کرده‌‌ده - نقده، کوه سهند - چهاربورج و کوه سهند - روستای کرده‌‌ده، سایر مقایسۀ جمعیت‌‌ها با هم اختلاف معنی‌‌دار نشان نداد (جدول 3).

خوشه‌بندی یا تحلیل خوشه‌ای (Cluster Analysis)

خوشه‌بندی یا تحلیل خوشه‌ای فرآیند گروه‌بندی افراد با ویژگی‌های مشابه یا اندازۀ متغیر مشابه است. همان‌‌گونه که در شکل 6 مشاهده می‌‌شود، در خوشه‌‌بندی جمعیت‌‌ها به‌‌واسطۀ بال جلو (تصویر بالا)، جمعیت‌‌های نقده، کوه سهند، روستای کرده‌‌ده و ملکان یک خوشه و جمعیت‌‌های میاندوآب و چهاربورج نیز هرکدام یک خوشۀ مجزا تشکیل داد؛ همچنین خوشه‌‌بندی جمعیت‌‌ها برحسب تشابه در بال عقب سه خوشۀ مجزا را نشان داد. جمعیت‌‌های نقده و ملکان یک خوشه، جمعیت‌‌های میاندوآب، کوه سهند و روستای کرده‌‌ده یک خوشه و جمعیت چهاربورج نیز یک خوشه را تشکیل داد.

رابطۀ بین تغییر اندازه و شکل بال، آلومتری یا ایزومتری

رابطۀ بین تغییرات اندازه و تغییرات شکل بال‌‌ها با رگرسیون بررسی شد. نتایج تجزیه و تحلیل آماری نشان داد که تغییرات آلومتریک در بال جلو وجود داشته است (Wilk’s Lambda = 0.79237403, P <0.05)؛ ولی در بال عقب دیده نشد و این مسئله به این معنی است که تغییرات اندازه و تغییرات شکل در بال جلو به یکدیگر وابستگی دارند.

بحث

زنبور عسل به‌‌دلیل نقشی که در گرده‌‌افشانی گیاهان و محصولات گلدار و تولید عسل دارد، یکی از مهم‌‌ترین حشرات است. اندازۀ بازار جهانی عسل در سال 2021 حدود 17/8 میلیارد دلار بود و انتظار می‌‌رود تا سال 2029 به 69/12 میلیارد دلار افزایش یابد (Fortune Business Insights, 2022). تخمین زده می‌‌شود که زنبور عسل مسئول 12 از 16 میلیارد دلار ارزش گرده‌‌افشانی سالانه در ایالات متحده است (Calderone, 2012; Rader et al., 2016; Khalifa et al., 2021). کارشناسان کاهش چشمگیر سالانه در کلنی‌‌های زنبور عسل از سال 2006 را گزارش کرده‌‌اند. این کاهش به دلایل زیادی مانند آفات، آفت‌‌کش‌‌ها، بیماری‌‌ها، از دست دادن زیستگاه، کاهش گونه‌‌ها و تنوع ژنتیکی نسبت داده شده است (Nowierski, 2021). تنوع ژنتیکی گونه به آن کمک می‌‌کند تا بهتر با شرایط محیطی و بیماری‌‌های متغیر مقابله کند (Somero, 2012; Forsman, 2014; Dillon and Lozier, 2019)؛ بنابراین نظارت بر تنوع و حفظ آن دارای اهمیت است. تنوع فنوتیپی گاهی اوقات بیانگر تنوع ژنتیکی است (Crispo, 2008; Silva et al., 2013)؛ بنابراین در این پژوهش از تنوع ساختار بال برای ارزیابی تنوع جمعیت زنبور عسل در مناطق مطالعه‌‌شده استفاده شد. منطقی است که فرض شود شیوه‌های زنبورداری مانند جابجایی کلنی‌های زنبور عسل و تجارت زنبور عسل ممکن است به‌‌تدریج تنوع زنبورها را کاهش دهد؛ بنابراین فرض بر این است که تنوع در جمعیت‌های زنبور در مکان‌های مطالعه‌‌شده وجود ندارد.

در این پژوهش، از اندازۀ مرکزی بال‌‌ها به عنوان معیاری برای بررسی تنوع اندازه در بین جمعیت زنبور عسل استفاده شد. میانگین اندازۀ مرکزی بال جلو و عقب نشان داد جمعیت زنبور عسل چهاربورج بزرگ‌‌ترین و جمعیت زنبور عسل ملکان کوچک‌‌ترین بال جلو و عقب را دارند. تحلیل ANOVA و تحلیل تعقیبی post hoc نشان داد در بال‌‌های جلویی نه تا از 15 مقایسه دوبه‌‌دو و در بال عقبی نه تا از 10 مقایسه دوبه‌‌دو دارای اختلاف معنی‌‌دار است و در این زمینه بال جلو و بال عقب به‌‌طور دقیق الگوی یکسانی نشان می‌‌دهند؛ همچنین از مختصات لندمارک انطباق‌‌یافته برای ارزیابی تفاوت معنی‌دار در شکل بال با استفاده از تحلیل‌های MANOVA و تحلیل تعقیبی post hoc استفاده شد. داده‌‌های به‌‌دست‌‌آمده نشان داد در بال‌‌های جلویی 14 تا از 15 مقایسه دوبه‌‌دو و در بال عقبی سه تا از 15 مقایسه دوبه‌‌دو معنی‌‌دار است. با نگاه دقیق‌تر به داده‌ها، در رابطه با اندازۀ مرکزی جمعیت‌‌های آذربایجان شرقی و آذربایجان غربی باوجود نزدیکی برخی ایستگاهها با یکدیگر، تفاوت معنی‌‌دار نشان دادند. در رابطه با شکل بال نیز جمعیت‌‌های تمام ایستگاههای مطالعاتی به‌‌جز ایستگاههای کوه سهند و کرده‌‌ده با هم اختلاف معنی‌‌دار داشتند؛ اما در شکل بال عقب چنین الگویی مشاهده نشد و تنها جمعیت‌‌های کوه سهند با کرده‌‌ده، کوه سهند با چهاربورج و کرده‌‌ده با نقده اختلاف معنی‌‌دار نشان دادند. تئوری جداسازی با فاصله (IBD) بیان می‌کند که وقتی پراکندگی (یعنی جریان ژن) بین مناطق یا جمعیت‌ها ازنظر جغرافیایی محدود می‌شود، تفاوت‌های ژنتیکی ممکن است به‌‌صورت محلی جمع شود (Slatkin, 1993; Wright, 1943) و نظریۀ جداسازی توسط محیط (IBE) بیان می‌کند که تفاوت‌های ژنتیکی با تفاوت در محیط زیست افزایش می‌‌یابد (Wang and Summers, 2010; Sexton et al., 2014; Wang & Bradburd, 2014)؛ همچنین داده‌‌های به‌‌دست‌‌آمده در پژوهش حاضر نشان می‌‌دهد که تغییرات در اندازۀ بال و تغییرات در شکل بال در بین جمعیت‌‌ها در بال جلو وابسته به هم و در بال عقب مستقل از یکدیگر است.

امکان مطالعۀ خصوصیات مورفولوژیکی و مولکولی برای ارزیابی تمایز بین جمعیت‌‌ها وجود دارد (Dutech et al., 2005). مورفولوژی بال ارزیابی‌‌شده با روش مورفومتریک هندسی برای شناسایی و تمایز گونه‌های مختلف (Santoso et al., 2018; Ndungu et al., 2023)، زیرگونه‌ها (Henriques et al., 2020; Abed et al., 2021) و جمعیت‌های (Boulhasani, et al., 2018; Aglagane et al., 2022; Buala & Sopaladawan, 2022) زنبور عسل استفاده شده است. هوش مصنوعی، نرم‌‌افزارها و برنامه‌‌های کاربردی مبتنی‌‌بر وب نیز به‌‌منظور تسهیل استفاده از ریخت‌‌شناسی بال برای طبقه‌‌بندی و تجزیه و تحلیل بیشتر زنبورهای عسل توسعه یافته‌‌اند (Bustamante et al., 2021; Rebelo et al., 2021; Rodrigues et al., 2022). برای ارزیابی تنوع جمعیت‌های زنبور عسل در چند نقطه از استان‌های آذربایجان شرقی و غربی از روش مورفومتریک هندسی استفاده شد. به‌‌طور کلی، داده‌های به‌‌دست‌‌آمده نشان می‌دهد که با وجود اینکه تمام ایستگاههای مطالعه‌‌شده در کمتر از 12500 کیلومتر مربع واقع شده‌اند، تنوع چشمگیری در اندازۀ بال و شکل بال در هر دوی بال‌های جلویی و عقبی وجود دارد که تنوع جمعیت‌های زنبور عسل را در این منطقه نشان می‌دهد. مطالعات نشان می‌‌دهد که تنوع ژنتیکی، سلامت کلی کلنی‌‌های زنبور عسل را بهبود می‌‌بخشد. کندوهای دارای تنوع ژنتیکی، مقاومت بیشتری نسبت به بیماری دارند و به محرک‌‌های محیطی مختلف پاسخ مؤثرتری می‌‌دهند. فقدان تنوع ژنتیکی برای زنبورهای عسل، آسیب‌‌پذیری ایجاد می‌‌کند؛ به طوری که زنده نمی‌‌مانند در اقلیم درحال تغییر که اکنون مرطوب یا خشک‌‌تر از حد معمول است؛ همچنین نگرانی وجود دارد که ناتوانی زنبور عسل در مبارزه با بیماری یا عفونت انگلی ممکن است بر پایداری زنبورداری تأثیر منفی بگذارد (Tarpy et al., 2013; Cridland et al., 2018; Alburaki et al., 2023).
